sometimes there are holes for part of the casting process but are usually sealed/plugged before the final firing.
It is very unusual to see a hole filled /sealed with caulk.

Just grab hold of the part on the end of the threaded nipple with some pliers and take it off. That threaded nipple is only holding the escutcheon on.
