How long it usually take for a complete sample preparation?

How long it usually take for complete sample preparation when using an HPLC column?

So far as I know, sample preparation is a time-consuming process, amino acid HPLC column can only take 30 min for this process. Okay, hope the administrator can warm me instead of banning me id he/she notice this self-questioning.

Some advertise as little as 10 minutes.
What are the typical reasons this would be required?(From A plumbing standpoint.)
